Using Sperm Transcriptome in Search for Novel Biomarkers of Male Infertility

Currently infertility affects 10 to 15 percent of couples. In nearly 50% of all cases male factor contributes to infertility of the couple. Majority of causes of male infertility remains unexplained. For this reason, finding simple and clinically useful tools for improving male infertility diagnostics can be important at present. Based on the analysis of sperm transcriptome, in this diploma thesis we aimed to find genes that show differential expression between normal and pathological sperm, this could provide information about molecular basis of male infertility, moreover, expression profile of such genes in sperm could be employed for noninvasive diagnosis of male infertility. This study was conducted by using 67 sperm samples, including 16 control sperm samples from fertile men. The samples of infertile patients were divided into 3 groups according to morphology and motility using results of standard sperm evaluation according to WHO. Molecular genetic research has a very dynamic development, and also genetically based economic production traits of domestic animals and plants are in the spotlight of researchers and breeders. This paper summarizes an actual overview on the field of candidate genes and their SNP associations with beef quality and yield, with their short description, resulting assigning the genes for practical breeding. There were described 10 genes: CAST (calpastatin), PLAG1 (pleomorphic adenoma gene), ANK1 (ankyrin 1), FABP4 (fatty acid binding protein 4), FASN (fatty acid synthase), SCD (stearoyl-CoA desaturase), LEP (leptin), MSTN (myostatin), IGF2 (insulin-like growth factor 2) and TG (thyroglobulin).
The analysis of chosen candidate loci influencing commercial properties and animal health

The aim of this thesis was to analyze the polymorphism in position -371bp (related to ATG start codone) of MSTN gene, SNP in position 1984bp of MYF5 gene and influence of these polymorphisms on tenderness, water holding capacity, pH and color of meat. Samples (241) of bull meat of Czech pied cattle were used for analysis. PCR-RFLP method was applied to genotype MSTN and MYF5 genes. For polymorphisms detection was used restriction endonucleases DraI (for MSTN gene) and TaqI (for MYF5 gene). From established genotypes were computed their frequencies and alleles frequencies. The frequencies of genotypes in MSTN gene were AA=0,729, AB=0,258, BB=0,013 and alleles frequencies were A=0,858, B=0,142. The frequencies of genotypes in MYF5 gene were AA=0,181, AB=0,542, BB=0,278 and alleles frequencies were A=0,452 a B=0,548. Using statistical analysis, the influence of genotypes of MYF5 gene on the water holding capacity, pH and the color of meat was determined.
Effect of selected candidate loci on meat quality indicators by cattle

This study was aimed on analysis of polymorphism at position 2141 in exon 5 of the growth hormone gene (GH) and polymorphism at position 257 in exon 10 of the growth hormone receptor gene (GHR) and to determine the influence of these polymorphisms on meat tenderness. To analysis has been used 333 meat samples of Czech Pied bulls. Genotypization of GH and GHR loci was performed by PCR-RFLP. For detection of single nucleotide polymorphism in both genes was used restriction endonuclease AluI. To determine meat tenderness was used the method of measuring shear force by Warner and Bratzler. The shear force values of raw meat samples were measured at day 14 post mortem. Then was statistically evaluated the association between genotypes and shear force. In the study population occurred for GH locus 166 individuals with genotype LL, 161 heterozygotes LV and 6 homozygotes VV. The relative genotype frequencies were thus 0.499 (LL), 0.483 (LV) and 0.018 (VV). The frequency of L allele was 0,74 and of V allele was 0,26. For GHR locus was found in the study population 178 homozygotes AA, 105 heterozygotes AG and 50 homozygotes GG. The relative frequencies of genotypes were 0.535 (AA), 0.315 (AG) and 0.15 (GG). The frequency of A allele was 0,692 and of G allele was 0,308. Using statistical analysis revealed a significant effect of genotype GH gene on shear force, tenderness resp. (P<0,05). For GHR locus showed no effect of genotype on shear force (P>0,05).
Cattle Candidate Genes for Meat Production Traits

The objective of this study was to compile a summary of the most important candidate genes for meat production. The studied genes were: GH, GHR, MSTN, MyoD family, leptin, IGF, TG5, SCD, DGAT and STAT5A. Growth hormone (GH) is involved in physiological processes of growth and metabolism. Growth hormone receptor (GHR) has been proposed as a candidate gene for meat production in cattle. Myostatin is a significant marker. It affects the amount of muscle, reduces marbling and elevate meat tenderness. MyoD family are proteins that play a role in regulating muscle differentiation. MyoD family include the genes: MYF3, MYF4, MYF5 and MYF6. Leptin is in association with the storage of fat, food intake and energy balance. The thyroglobulin gene (TG5) is the precursor for thyroid hormones. These hormones have important role in formation and differentitation of cells. Product of the SCD genes is stearoyl? CoA denaturase. This enzym is responsible for conversion of saturated fatty acids into monoinsaturated fatty acids. DGAT1 genes is a candidate gene for marbling of meat and fat in milk. STAT5A gene is a group of transcription factors and is very important intracellular mediator of prolactin.
Cattle Candidate Genes for Milk Production Traits
